Abstract : There is an urgent need for cancer therapies with limited side effects, and silver nanoparticles (AgNPs) have shown to be promising in this regard. In spite of this, improved environmentally friendly methods are required for the preparation of AgNPs. This study used Juniperus procera (J. procera) and Azadirachta indica (A. indica) extracts, medicinal plants widely used for treating a variety of ailments, to biologically synthesize AgNPs. Bioreduction of silver ions (Ag+) into silver nanoparticles (Ag0) is facilitated by plant phytochemicals such as flavonoids, tannins, and polyphenols. The shape, size, morphology, and optical properties of silver nanoparticles significantly affect their chemical, physical, and biological activities. The study, therefore, has also optimized the biosynthesis conditions, as a first attempt for the fruits and leaves extract of J. procera and A. indica fruits extract, to produce silver nanoparticles with precise morphologies. The biosynthesized AgNPs were characterized using a variety of techniques. Additionally, this study is the first to examine the cytotoxicity of green synthesized AgNPs using extracts of J. procera and A. indica against human lung cancer cell lines (A549 and H1975). In all extracts, UV-Visible spectroscopy detected surface plasmon resonance bands for biosynthesized AgNPs within the typical AgNPs range (400-500 nm). In the morphology evaluation of AgNPs, the nanoparticles (NPs) were found to be spherical with diameters between 14 and 18 nm. A dynamic light scattering (DLS) analysis revealed that all green synthesized AgNPs exhibited a diameter between 100 and 300 nm. The colloidal stability of AgNPs was between -23 and -35 mV according to a measurement of zeta potential. Fourier transform infrared spectroscopy (FTIR) revealed that AgNPs surfaces are embedded with biomolecules, such as polyphenols and flavonoids. Synthesized AgNPs exhibited different levels of toxicity against lung cancer cells A549 and H1975. Moreover, this study showed stable AgNPs with an acceptable morphology and high yield, which can be used for a variety of medicinal purposes. As a conclusion, AgNPs produced from the A. indica fruit extract have the highest toxicity against A549 white IC50 of 64.46 µg/mL.
