Abstract : Chitin and chitinolytic enzymes are gaining importance for their biotechnological applications. Chitinases are enzymes that degrade chitin and contribute to the generation of carbon and nitrogen in the ecosystem. Microorganisms are producing Chitinases, which can inhibit the growth of many fungal diseases that pose a serious threat to global crop production. Currently, efforts are being made to discover producers of chitinolytic enzymes that degrade chitin. The aim of this study was production and characterization of thermo-tolerant Chitinase from bacteria and actenomycets, isolated from western region, Saudi Arabia. Colloidal chitin from shrimp shells was prepared and used for isolation of chitinolytic bacteria on Mineral chitin agar medium from different sources. The most active isolates were AMM1 which was characterized and identified as Alcaligenes aquatilis and AMB8 which was identified as Kitasatosporia sp. The identification was confirmed using 16SrRNA. For Alcaligenes aquatilis, maximum enzyme production was carried out at 35°C, using Mineral chitin broth medium B (pH7.0), containing yeast extract (5 g/l), Glucose 2.5 (g/l), chitin (15 g/l) and inoculated with 6x106 CFU/ml. For Kitasatosporia sp., maximum enzyme production was at 30°C, using Mineral chitin broth medium B (pH 7.0), containing yeast extract (5 g/l) and Glucose 2.5 (g/l), chitin (10 g/l) and inoculated with 4x104 CFU/ml. The enzyme was extracted, purified and characterized. Maximum activity of the pure bacterial enzyme was at 35ºC, pH 7, with enzyme concentration of 3% and substrate concentration of 0.6. For the enzyme of Kitasatosporia sp., maximum activity was at 30ºC, pH 7, enzyme concentration of 3% and substrate concentration of 0.6 %. It was found that the two tested enzymes were affected by the presence of metal ions. The molecular weight of the two tested Chitinases were determined. The obtained Chitinases were used to inhibit spore germiatiom and fugal growth of 6 tested fungi, thus it can be used in biocontrol process.
